Abstract
T. cacao husks or pod from regular and black pod infected sources were analyzed and compared for their total soluble protein and protein bands using gel electrophoresis. The study tested different extraction media and protocols to increase the extraction of cacao proteins from the husks and leaves to ultimately qualify and quantify the proteins. 8 different extraction buffers and several extraction protocols were tested. The resultant extracted proteins were then subjected to Gel electrophoresis to compare the differentiated protein bands between the healthy and infected husks. Furthermore, protein quantification method using Bradford method and silver stain method were conducted. The result of the study suggests that there is reduction of total proteins from the infected cacao husks when compared to the healthy counterpart. Furthermore, some protein bands were noticeably different for both the infected and healthy cacao pods. This extraction method will be useful for further analyses for comparing healthy and infected T. cacao husks proteins specially when using chromatographic techniques.
